Abstract

This master thesis has two major parts, on the one hand, already existing Physcomitrella patens PIN-LIKES (PILS) mutants were analyzed, to deepen the current understanding of their role auxin transport. On the other hand, novel mutants of PILS proteins in P. patens were generated. With respect to the first objective, special attention was payed to monitoring the growth and phenotype of the mutants in different environments. To this end, the protonemal tissue of the mutant lines was grown in media with either an auxin biosynthesis inhibitor (Kyn) or different natural (IAA) and synthetic auxins (NAA). In addition, high temperature studies were carried out, growing mutant protonema in a standard medium different temperatures: 21°C, 25ºC, 29ºC and 35ºC. As for the second objective, stable transgenic lines of P. patens were generated that will be used for localization and overexpression studies. The moss was transformed with plasmid constructs previously generated in the laboratory.
