Abstract

Mesenchymal Stem Cells (MSCs) are multipotent cells considered as excellent candidates for different immunomodulator therapies. Several evidences suggest that their immunomodulatory properties could be dependent on the inflammatory environment. Macrophages are cells of the immune system that participate in the development and resolution of inflammation. They can adopt a pro-inflammatory (M1) or anti-inflammatory (M2) phenotype. The aim of this work was to evaluate the effects on macrophage activity by soluble factors produced by MSCs derived from human decidua (DMSCs) cultured in pro- or anti-inflammatory conditions. RAW 264.7 cells were subjected to polarization towards M1 and M2 phenotypes in the presence of DMSC secretomes produced in different cytokine environments propitiated by IL-6, IFN-γ, TGF-β or IL-10. Gene expression levels of M1 and M2 markers were quantified by RT-qPCR. On the other hand, we analyzed the endocytic capacity of M1 cells subjected to the different DMSC secretomes by flow cytometry. The results showed that the power of the DMSC secretome obtained under a neutral medium to modulate the expression of M1 (IL-1β and IL-6) and M2 (Arg1) markers is intensified by DMSC pretreatment with IL-6, IFN- or IL-10. On the contrary, TGF- treatment reduces such quality of the soluble mediators produced by DMSCs. We also found differences among the pre-treatments of DMSCs assayed in terms of their influence on the endocytic levels of M1 macrophages. These results demonstrate the influence of the inflammatory conditions on the action of DMSCs and can be helpful in understanding the diversity of the roles of these cells.
